Prucalopride
Description Prucalopride is a medication used to treat chronic idiopathic constipation (CIC). It is a first-in-class dihydrobenzofuran-carboxamide compound. It is a selective and high-affinity serotonin 5-HT4 receptor agonist.
Indication Prucalopride is indicated for the symptomatic treatment of chronic constipation in adults, particularly in cases where laxatives have failed to provide adequate relief.
Mechanism of Action Prucalopride acts as a selective agonist for serotonin type 4 (5-HT4) receptors. By binding to and activating these receptors in the gastrointestinal tract, it stimulates colonic motility. This leads to the release of acetylcholine, an excitatory neurotransmitter that causes the contraction of the muscle layers of the colon. This action enhances the natural propulsive movements of the colon, known as peristalsis, which increases bowel motility and facilitates defecation.
